Stacking gel and sample buffer for SDS-PAGE.

Tris base	12.11 g	60.57 g	121.1 g
HCl (12 M)approximate; finish with a pH meter	7.909 mL	39.54 mL	79.09 mL
Final volume with water	100 mL	500 mL	1 L

Dissolve Tris base in about 80 % of the final volume of purified water.
Adjust to pH 6.8 at 25 °C with HCl (12 M), adding slowly while stirring.
Make up to the final volume with water.
Use fresh or store at 4 °C for up to a month.

Target pH is more than 1 unit from pKa (8.07); buffer capacity will be low.
